Combination of fluvastatin and niacinamide is synergistic against development of type 2 diabetes mellitus in Wistar rats.
The effect of a combination of fluvastatin (FLV), an antihyperlipidemic agent, and niacinamide (NIA), an antithyroid drug, was assessed on 2-wk- and 12-wk-old rats (n = 5). Two series of experiments were performed. In Experiment 1, the rats were treated with FLV, NIA, and combinations of FLV+NIA (FLV/NIA) during the last 2 wk of their life span to compare the effects of the combination with those of FLV and NIA alone. In Experiment 2, rats were treated with various regimens starting from age 2 wk for 12 wk to evaluate the synergistic effect of FLV+NIA and FLV+NIA+FLV on the development of type 2 diabetes mellitus. At the end of the treatment, the rats were killed for biochemical and histological evaluations. A combination of FLV (30 mg/kg body wt) and NIA (20 mg/kg body wt) showed synergistic effects on total cholesterol (TC), triglyceride (TG), phospholipid (PL), and low-density-lipoprotein cholesterol (LDL-C) levels. Analysis of these lipid parameters revealed that the combination gave a better effect than each drug alone. There was no difference in the TC, TG, and PL levels between groups given FLV (10 mg/kg body wt) and those given NIA (10 mg/kg body wt) and FLV (10 mg/kg body wt)+NIA (10 mg/kg body wt), but the LDL-C level was lower in the latter group than in the former. Similar results were obtained in Experiment 2. Administration of FLV (20 mg/kg body wt), NIA (10 mg/kg body wt), and FLV+NIA, as well as FLV+NIA+FLV, for 12 wk starting at age 2 wk, prevented the occurrence of type 2 diabetes mellitus, as judged by blood glucose levels and the microangiopathic changes observed in the aortas of the rats.
